Target note

Full name: Integrins αvβ3 and αvβ6
Nature: Adhesion receptors for the extracellular matrix that recognise the RGD motif.
Normal expression: αvβ3: endothelial cells in neoangiogenesis, osteoclasts. αvβ6: almost absent from healthy adult epithelium.
Disease expression: αvβ3: tumor neovessels, melanoma, glioma. αvβ6: carcinomas of the pancreas, head and neck, lung, colon; also in fibrosis.
Value: αvβ6 offers high contrast in cancers that lack validated targets, such as the pancreas.
Approaches: RGD peptides (3PRGD2, EB-RGD), αvβ6-specific peptides (5G, ABM-5G); ¹⁷⁷Lu.
Indications explored: Pancreatic adenocarcinoma, head and neck and lung cancers, solid tumors (phase 1).
Points of caution: Renal and intestinal uptake; αvβ6 is also expressed in fibrosis.
