Target note

Full name: Glypican-3
Nature: Membrane-anchored (GPI) heparan sulfate proteoglycan, oncofetal.
Normal expression: Fetal liver; absent from healthy adult liver.
Disease expression: Hepatocellular carcinoma (about 70 to 80%), hepatoblastoma.
Value: High contrast between tumor and healthy liver, in a cancer where other targets are limited.
Approaches: Vector labeled with ²²⁵Ac (RYZ801), paired with an imaging agent (RYZ811).
Indications explored: Unresectable GPC3-positive hepatocellular carcinoma (phases 1-2).
Points of caution: Underlying liver function often impaired (cirrhosis).
